human culture-expanded activated autologous T lymphocytes for cell-based immunotherapy. The cells are mechanically and enzymatically isolated from resected tumour biopsy of patients with metastatic melanoma and cultured in media containing recombinant interleukin 2 (IL-2) and foetal bovine serum (FBS). The cells are activated using an anti- CD3 antibody and irradiated allogeneic peripheral- blood mononuclear cells (PBMCs) in media supplemented with human AB donor derived serum and IL-2. The T lymphocytes (>80%) are a heterogeneous mixture of CD4+ and CD8+ tumour- infiltrating lymphocytes (TIL) predominantly with effector memory and central memory cell subsets. The T lymphocytes demonstrate the ability to kill and impede tumour growth in a co-culture based cytotoxicity assay.
