autologous mesenchymal progenitor cells prepared from subcutaneous adipose tissue of patients with osteoarthritis (40-75 years old), collected by liposuction. The adipose tissue is digested and the released cells placed in culture medium containing insulin-transferrin-selenium, recombinant human basic fibroblast growth factor (bFGF), and recombinant human epidermal growth factor (EGF). The cells are frozen, resuscitated and culture expanded to around passage 4-5. The cells express mesenchymal progenitor cell marker CD73, CD90, and CD105 (>95%) and do not express human leukocyte antigen (HLA) DR, CD14, and CD45 (<2%). The cells are capable to differentiate into chondroblasts under standard in vitro tissue culture-differentiating conditions and demonstrate the inhibitory effect of mesenchymal progenitor cells on the proliferation of T lymphocytes.
