allogeneic human retinal progenitor cells (hRPCs) derived from foetal retina (17-20 weeks gestation). The retinal tissue undergoes dissection and digestion and the obtained cells are grown on fibronectin-coated plates under hypoxic conditions in serum free high nutrient media. The final cell population is characterized by the expression of retinal specification genes (nestin, >95%; Sox2, >85%; Ki67, >50%) and as well as by evidence of differentiation into neurons (neuronal migration protein doublecortin (DCX)) and glia (glial fibrillary acidic protein (GFAP; <5%). The cells secrete osteopontin (OPN), and the secreted OPN together with other trophic factors can give protection to a human retinoblastoma (hRB) cell line treated with a histone deacetylase inhibitor.
