ecombinant, replication-incompetent E1-deleted adenovirus serotype 26 vector encoding a full-length pre-fusion stabilised severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike (S) glycoprotein (sequence based upon the original Wuhan isolate of 2019, whole genome sequence NC_045512), with two amino acid changes in the S1/S2 junction (R682S and R685G) that knock out the furin cleavage site, and two proline substitutions in the hinge region (K986P and V987P). Expression of the transgene is under control of a synthetic human cytomegalovirus promoter with a tetracycline operon (TetO) sequence inserted upstream of the transgene and the simian virus 40 (SV40) polyadenylation (polyA) signal.
