recombinant replication-competent oncolytic vaccinia virus (VACV; strain Copenhagen) encoding three codon-optimised transgenes: (i) anti-cytotoxic T- lymphocyte-associated protein 4 (CTLA-4) immunoglobulin G1 γ1-chain fused via a synthetic T2A peptide with human immunoglobulin kappa chain (anti- CTL4 antibody) under control of the VACV H5R promoter, (ii) human Fms-like receptor tyrosine kinase 3 ligand (FLT3L), isoform X7, under control of the VACV B19R promoter, and (iii) membrane bound human interleukin 12 subunit α (IL-12A, p35) under control of a synthetic VACV late promoter (pLate). The virus carries major genomic deletions at the 5' end (CopMD5p) and 3' end (CopMD3p) of the genome with the anti-hCTLA-4 transgene inserted into the CopMD5p region. In addition, insertion of the FLT3L and IL-12p35 transgenes leads to disruption of the B8R locus.
